Is it ok to say - "Do the needful" ?

'needful' being an adjective, can we use it in the above manner?

It may be standard where British English prevails, but it sounds odd to American ears. See Required Modern Sentence and the discussion following for various opinions. Personally, I agree with those who think it sounds like a euphemism for some bodily function. It would be best to avoid it
