Summary:
This paper explained the relationship between telomeres length and age, which is associated with genomic instability and high risk of cancer. The authors studied telomeres length in normal colorectal tissues, normal epithelium adjacent to or distant from colon adenomas and neoplasm’s, Peripheral blood lymphocytes (PBL), and Different lesion size of adenoma and cancer tissues (>2 cm, 1-2 cm, and <1 cm). They did their study in a wide verity of patients in different ages range (0-93 years) and from both sex.
The analysis of telomeres length was done by using two techniques, Q-Fish and Q-PCR, to measure telomeres length in epithelial, stroma tissues, and small amount of DNA. The cells were extracted and isolated from frozen biopsies using a shake-off method. In addition, the comparison and the analysis of telomere length in different ages and tissues were seen in simple linear regression and two-segment model.
The results from Q-Fish showed that telomeres in the epithelial and stroma of colon in young person are longer that old person, therefore, by measuring the epithelial/stroma telomere ration of different set of ages, they found that a decrease of telomere length is related to increase age except in the age of 80 to over 90 the telomere start to increase. They believed that the telomerase enzyme function is reactivated over the age of 80.
Moreover, the results given by Q-PCR, which measured the telomeres length in the isolated epithelium, stroma, and PBL, showed that telomeres length in epithelium and PBL are decrease by age, however telomeres lengths in stroma did not show any association with age. This finding proved that individuals of age 60 and over with short telomeres are in high risk of mortality.
Interestingly, the results giving by Q-Fish for telomere length in adenoma with different lesion size (<1, 1-2, >2 cm), colorectal cancer and normal mucosa near adenomas and cancer showed that, in >2 cm adenoma, telomeres length are shorter than normal adjacent and distant tissues, however, no difference in telomere length in small lesion adenoma and cancer tissues. These results reflect the genomic instability in short telomere that may affect the malignant transformation of large ademonas and telomerase reactivation in colorectal cancer cells that prevent telomere length reduction.
This paper included smoking and nonsteroidal anti-inflammatory medications (NSAID) effects in telomere length. Q-PCR showed no relation between telomere length and gastrointestinal disease or NSAID. However, smoker had shorter telomere length than nonsmoker, due to tobacco genotoxins affect on telomere.
This study suggested that telomeres length in colon may differ from other tissues and this may related to cell proliferation rate, telomerase enzyme activity in stem cells, genotoxic exposure history, and genomic instability.
Critique:
Short paper with a direct title, full of information and results, it was easy to read as well as the purpose that achieved clearly throughout the paper. The introduction covered the main points in a simple way. The materials or samples were collected from a wide verity of people from different ages which made the result more convenient and believable. However, the methods part was not fully described, I had to look up in previous papers to know how some tests have been performed and applied. For example, intra and inter assay and statistical analysis. Figures and tables were clear, well described and easy to understand. Overall, the paper does a good job in pointing out that a lot is done towards analyzing the telomeres length in different body parts and in different ages.
