Actually ,my question was not clear.

I want to know whether "I need you to hold the fort" means "I need you for holding the fort" or "I need for you to hold the fort".

Will a comma disambiguate the sentence if I write it as "I need you , to hold the fort" ?

I don't think the comma helps, although it might push us toward your first interpretation.
In actual fact, I'm quite sure the typical native speaker would take the second interpretation.
