No, it would not be natural. Presumably you mean, ‘Please put just a little sugar in my coffee.’
When sugar was in cubes rather than in paper packets, "Just one lump of sugar, please."

Yes. I have friends that visit me from time to time and if I make coffee for them I ask "How much sugar? One teaspoon or two? Or do you want to do your own?" If they let me do it, I add the sugar for them and serve them their coffee. It's all very informal.
