The definite article or not?
She was sitting on a/the chair in my room when I asked her if she want to be my new girlfriend.
(there were 4 chairs in my room)
You have more than one chair in your room, so "the", which indicates that the reference is unique in the expression "the chair", is not possible. Use "a chair" in your sentence. Note: Your sentence is in the past ( was sitting, asked ), so you need 'wanted' instead of 'want'.

"the chair" would sound as if there was only one chair in your room.
You could also eliminate "on a chair" entirely without a great loss.
