I'm not that big of a loser.
I'm not that much of a loser.

Is one of the 2 correct and natural?

"that big of a ..." is incorrect in standard English (to me in the UK it sounds totally wrong, but it seems fairly widespread in the US). In standard English it should be "that big a ...".

Otherwise, there is not a great difference in meaning in the contexts I can envisage.

It is definitely informal but quite idiomatic here in the states.
