Make no mistake that I'm not your friend. If I had a chance, I'd make your life a living hell.

Your first sentence should be broken in two rather than connected by the word 'that':
- Make no mistake. I'm not your friend.

I'd probably use 'the chance' rather than 'a chance' in your second sentence, but that's just my personal preference.

How about the following?

It's time we wake up and move our country forward. Make no mistake. We're among the poorest countries in the world!

I don't feel comfortable with the order of the sentences. In this case, it seems to me it would be better this way:
- Make no mistake. We're among the poorest countries in the world! It's time we wake up and move our country forward.

Some might prefer your "it's time" sentence worded this way:
- It's time we woke up and moved our country forward.
