What is the different between...

" Is he dead ?" and " Did he die ?".

There's not a lot of difference.

Is he dead? Focuses a bit on the present.
eg Tom owes me $100. Is he dead?

Did she die? Focuses a little more on the past.
eg I know Mary was very ill in 2010. Did she die?
