1) Should I say "He lives in the South" or "He lives on the South"?

2) Can I say "I had a class this afternoon"?

No to the first. The place is on the south side of the city, yes. Note, lower-case s and specifying the south of what place.

But "the South" (capital S) refers (when used here) to the southern part of the US, an entire region.
