Which one is correct, with 'in or of'?

The largest muscle in the human body is gluteus maximus.
The largest muscle of the human body is gluteus maximus.

I am not sure about using 'of', but I think it might be possible.

Both 'of' and ' in' are possible. But the latter is preferred.
Just a side thought....
As far as your sentence is concerned, I think 'biggest' muscle works idiomatically better than 'largest' in my opinion.
