You had better reply to this message ____ a couple of days.

The answer is "in" but I think that "after" is also okay to use, isn't it? Or is there a reason why "after" is not okay? Thank you so much as usual and have a good day.

"after" is certainly possible there but it would give the impression that you intentionally want to wait a couple of days before replying to that message (suggesting perhaps that you want to avoid something asked of you).
