No. There are two problems:

1. "first reason" as a noun phrase requires the definite article: "the first reason."

2. "the first reason" is a noun phrase. It cannot just "hang" in a sentence. It must serve in a legitimate nominal function such as subject, object, or appositive. So this is wrong:

X The first reason, my mother was born in Germany.

You need an adverb as the sentence introductory word.

For example:

There are two reasons. First, my mother was born in Germany.

Also prepositional phrases can be used:

For the first reason, I can tell you that my mother was born in Germany.
