in chemistry ? of chemistry?

"I took a course IN chemistry at a college." "I took a course OF chemistry at a college." I con't know which preposition I should use. Though I refered a sentence "I took a course in chemistry at a college."
Which one is the correct preposion IN or OF?

Thanks! I'll use "IN" between "course" and "chemistry" from now on, but please let me know why native speakers don't say "I took a course OF chemistry." Does it make different meanings?
