Can I say,

She is cooking at / in the kitchen.

She is cooking some food at the kitchen.

I think it depends on where the Kitchen is located.

If you are in the quarters (house, restaurant, etc) where the kitchen is, then, I would use 'She is cooking in the kitchen'.
If you are talking about a place, then I would use the latter, 'She is cooking at the kitchen on Broadway at 7th street.'
