I'm writing in regards to/with the work opportunity that I have come across on your website.


Actually, I'm not even sure if it sounds alright... especially the work opportunity part and I have come across...

Well, based on that piece of information, I'd say that the following versions are correct (at least as far as in/with regard to is concerned)

I'm writing in regard to the work opportunity that I have come across on your website.
I'm writing with regard to the work opportunity that I have come across on your website.

All three are fine, and I much prefer (and always use, if I must use any) your third one, 'regarding'.
In fact, I avoid and advise avoiding any of those forms when a simple 'about' will suffice.
