This is quite casual. It's appropriateness would depend on the type of work the job entails, and the sort of person who might be reading it.

Most Americans are well-acquainted with the expression. (Yes, it makes sense.)

My point is that if you need to learn the very foundational aspects of the job, what could you be bringing that would be useful to the employer?

A cover letter tells the employer why you would be helpful to them. What you want is pretty unimportant, and telling them you want to learn basics (plural) wouldn't inspire someone to want to hire you.
