What do you mean? = What is your intention?

I didn't mean to hurt you. = I didn't intend to hurt you.

I think it would be a little clearer with "for" added...i.e.,

Did you mean for John to post those letters?

BTW, in the U.S., it would be, "Did you mean for John to mail those letters?"
