1. What is the point?

"Point" in that sentence has the meaning of purpose/objective. You can ask "So, what's the point?" when you don't understand where someone is going with his/her story.

2. (The) point is....

This can be said in response to the above question or when the speaker/writer wants to clarify what has been previously said.

3. I mean it.

This can be said when you want to make sure that the person(s) you're talking to understand that you are serious about something you've said.

4. Knock it off. (= stop it; stop doing what you're doing)

edit: BTW, the plural persons is rarely used but it was convenient for me to just add the ending s.
