how to write a letter of application to a PHD program

I am applying for a PHD program at Delft University of technology. A letter of application is required. I am not an English native speaker and don’t have relevant experience before. I am writing here for help. What is the proper structure of the letter? What is most emphasized? Is there any reference letter?
