how to use "the"

"A railway line runs through the entire town from the northwest to the southeast."

vs

"A railway line runs through the entire town fromnorthwest tosoutheast."

1. Is it necessary to use "the"?
2. Is it grammatically correct if I capitalize Northwest and Southeast?

I would consider the to be optional here. There is no need to capitalize north-west and south-east , unless these are names of districts rather than directions. The names of directions such as north-west are usually hyphenated.
