For example, you know that Peter, John, and Andrew are playing over there. Someone says that her son is playing, and you ask, "Who is your son?" Because you already know the names of the boys, "John is my son." is okay.

I would say that under similar circumstances, when John is already known to the questioner, "Who is your son" could also be answered by "John is." or even just by the word "John."
