How does the placement of the word "can't" change the meanings, or does it?

I mean, does this two sentences mean the same???

1. Can't you give me a lift to school?
2. You can't give me a lift to school.

Or,

1. Can't I have a glass of water?
2. I can't have a glass of water.

It's difficult for me since i'm not a native speaker of english. So any help wud be appreciated....thnx
