Help with sentence

Which is correct to ask:

Do you know what is diabetes?
Or
Do you know what diabetes is?
I think that the first one is correct. Am I right?

Also, which is correct to say when I ask someone if they know about something (eg diabetes):

Do you know what 'it is' or 'is it'?

I am a little confused here. Please help.

What is diabestes?

Why is the 'is' before the diabetes in the second sentence? And do we use each statement?

Is the first sentence used when we directly ask someone about if they know about diabetes and the second is a more general question? I am confused!

Direct questions have a certain word order. What is diabetes?
Indirect questions have a different word order. ... what diabetes is.

Direct questions stand on their own. Indirect questions do not. Indirect questions often occur after introductory phrases like "I don't know", "Do you know", "I have no idea", "I wonder", "I can't imagine", "Please find out", etc.
