One of my friends says that natives don't say 'Do one thing'... this phrase is the typical of non-natives. Is this so?

No. Natives do not walk into a room and say, "Do one thing." and walk out.

They do say, for example:

If I could ask you to do one thing for me: keep an eye on Bobby while I'm away.
