Use firstly when talking about a list or enumeration.

e.g. Firstly, I would like to say that elephants are huge animals. Secondly, they don't eat humans. Thirdly, et cetera.

Use At first when talking about the beginning of something.

e.g. At first, John seemed pretty nice. But after a couple of months he revealed his true character.
