She reads in Grade 2. (seems very odd)
She is in Grade 2.

In America, we often say "second grade" or "2nd grade" instead of "grade 2."

So, you could say the following:

"She reads at a second grade level."
"She is in second grade."
"She studies in second grade."
"She studies second grade materials."

This is fine, but I wouldn't say it as a native English speaker in America. I would say 2nd grade.
