Sparkle always does the trick, does it not?
I'm sure that does it not is a sort of tag question but this is the first time I've come across the form ",does it not?"(the familiar one is ",doesn't it?") Are there any differences between the two? I assume that does it not sounds more formal or polite than doesn' because Rarity(the character saying the line) loves behaving in a posh way:) Thanks in advance!

I think it's a transcription error.

A common saying is;
That always does the trick, doesn't it?

"That" refers something that works very well. eg.
The lock is sticky and hard to open.
Try spraying a little graphite in the keyhole. That always does the trick!
