Disruption to the internal biological clock

Jet lag is caused by disruption to the internal biological clock, and may lead to digestive problems, tiredness, and sleep disruption.

Hi,

Is it also right to say "disruption of the internal biological clock" instead of "disruption to ... clock" in the above?

Besides, what on earth does "sleep disruption" refer to? Is it "totally sleepless?" Thanks.
