Difference between (.) and (b) in Phonetic transcription?

Hello guys!

I´m looking for an answer. Still can´t figure it out. Professor told me that there´s just one small difference which is so easy to spot that we
aren´t able to see it. He told me that it should be same in Spanish and English.

Those aren't written in IPA (the international phonetic alphabet), are they?

Can you translate them into IPA for us? Are you, by chance, referring to the difference between b and ß ?

Can you cite some English or Spanish words in which these occur?
