Dear

When sending an e-mail I usually start my message with "Dear,". My colleague however, always puts it in plural: "Dears,". I thought that it was never used in plural... Could somebody confirm this?

When writing the salutation of a letter, it is always Dear and never Dears.

In case you don't know, I'd like to clarify this a bit. You never say only the word 'Dear' as an opening. You need to say 'Dear + a name+ comma', eg

Dear Tom,

Dear Ms. Smith,

In my practice, emails are more casual, and I generally don't use a "greeting" Perhaps a 'hi', or something similar. But I'm not in a business where more formality might be appropriate.
