Hey, Jane. I need a big favor from you. My boyfriend is coming to town this weekend and I hope you can cover for me on Saturday. I already got Mike to cover my Sunday's shift.

Are there any grammatical mistakes? Also, are 'cover' and 'fill in' interchangable?

This is fine, but I would say "cover me on Sunday" or "cover my Sunday shift," rather than the apostrophe.

"Cover" and "fill in" mean roughly the same thing in this context, but aren't always interchangeable. Cover can be transitive or intransitive, but if an object is used, you want "cover" instead of "fill in." Fill in is something you do with a hole

Can you cover for me? Can you fill in for me?

Can you cover while I run get some cigarettes? Can you fill in while I run get some cigarettes?

Can you cover my shift? but not Can you fill in my shift? ("Can you fill my shift?" while a little odd, would be okay, I think)

Can you cover me tomorrow? but not Can you fill in me tomorrow?
