No, I'd think the oppostie without any futher context, that you had two yellow shirts and two red shirts.

I have two red-and-yellow shirts makes it clear you have two shirts, and each shirt has both colors.

If they were each one colour, I'd clarify it by saying I have two red shirts and two yellow shirts.

That's the reason I said "colors". People often have different pictures in minds when it comes to this kind of number, sizes and color context.

A: I like the stripe design on this shirt, especially the red and yellow, Oh here is one with red and yellow combination!

By the way I don't agree with GG. The picture this draws for me is that each shirt has both colours. And obviously it's not possible to clarify speach by adding hyphens. Anyway if she interprets it one way and I interpret it another it must be ambiguous.
