1/ In how many minutes will we get there by car?

These are better:

How long does it take to drive there?

3/ I gave you the least fill glass didn't fill your glass up because you are the designated driver.

hi
How many kilometers are on this car?

Is this the same How many miles does the car have on it? are they both correct?

Yes, that's fine.
Also - "What mileage does the car have?" "What is the odometer reading?"
