"Can't u..?" and "Can you...?"

Hi,

Would you please help me a little with 'interrogative sentences' and 'negative interrogative sentences'? In what situation should we use which? For example, what is the difference between 'Can't you do this for me?' and 'Can you do this for me?', please?

Thank you.
