I learned recently that "be up on something" means that somebody knows a lot about something. By the way, why do you say it? With what imagery do you use this expression? Do you mean "someone is in a high(er) position in knowledege about a certain matter"?
I'm a native English speaker and I have never heard of that expression meaning someone knows a lot about something. You can use the expression like this "the car is up to date" meaning the car is the newest version available. Or you could say "that girl is up to something" as in that girl is doing something shady or sneaky but you don't know exactly what she is "up to" yet.
