Articles of antecedents

1)Lee is an engineer who designed this machine.
2)Lee is the engineer who designed this machine.

#2 means the only engineer who designed this machine,so we should use "that" instead of "who"?

I'd like to know relevance between articles of antecedents and relative pronouns(who, which and that)
