Are you kidding me

Hi, I know there are two situations where people would say that line.

First one is said when someone told you something that you find it hard to believe because it's too good to be true or something like that.

My issue is with the second one.

You had a dinner with your colleagues including your boss after work. And your boss asks you personally if you would like a beer or coke, and your answer is,

"Are you kidding me? I'll take a beer any day"

I know that most non-native speakers will take this line the wrong way and be easily offended. But what about to native speakers? Would it still make you sound like you're being disrespectful to your boss?

And what does that line really mean in the 2nd situation?
