Applying for a job: resquesting info

1) Suppose I'm not sure whether I'd fit in the job being advertised as the info given on what the task implies is not clear enough. How should I handle this in my application letter? Should I write a request for more info on the letter too? Are there any samples of "requesting information letters" (I don't actually knwo if that is the technical term).

2) Suppose also that they are specifically asking for a postition that is not exactly your study field but you know it has some points in common with your spertise (sp?), how would you handle that? Are there any samples of application letters conveying this situation?

Thanks.
