I got three boxes of chocolate from See's Candies. I say to the cashier:

Yup, that's all.

OR

Yup, that's it.

My usual experience with cashiers is this.
I go to the counter and put my items on it.
She or he enters the amounts into the register and then says eg $36.40. please.

The question itself does not seem natural to me.

OK. My questions basically is if it's OK to say to the register:

That's it. OR That's all.

Like when you're at a drive thru and have completed your order.

Yes. It doesn't matter whether you say "That's it" or "That's all" when you are saying that there is nothing more. (You're finished. You have nothing more to say or to do.)

The same is true of the question form. "Is that it?" or "Is that all?"
