Can someone explain how to use "ain't" properly? because I confuse when I find "ain't nothing.." in a sentence.

As GPY explained, there is no correct way to explain and use "ain't " properly. It is a substandard way of saying " is not".
You probably heard one of these expressions before:

He ain't nothing = ( he is not anything important)
Ten thousand dollars ain't nothing
