There are a lot of people at the party.

"A lot of people" = plural, therefore "there are" .

There's a lot of people at the party. -- Nevertheless, this form is in common use and cannot be called incorrect, at least informally, for a couple of reasons: (1) It is easier to say and therefore more commonly used; (2) 'A lot of' is en route to becoming a quantifier.
